This chemo-enzymatic approach facilitates the coupling of synthetic, fully unprotected peptide fragments to each other and to (recombinant) proteins. The enzymes allow complete stereo- and regioselective ligation under aqueous conditions at near neutral pH. No recognition motifs are required. Enzyme efficiency and activity can be tailored using site-directed mutagenesis. The technology has been proven up to pilot plant scale for Exenatide, Thymosin-a1 and Liraglutide. It resulted in > 25% yield improvement compared to commercial scale standard SPPS. The technology has also been proven for the synthesis of (bio)conjugates, antibody-drug conjugates, interleukins and chemokines. Plasma proteins like C-reactive protein, mannose-binding lectin, glycoproteins and the like are also within reach of the technology.
